چکیده

background: malassezia are dimorphic, lipid-dependent yeasts, which are responsible for causing several cutaneous and systemic conditions. although cyclophilins (cyps) are highly conserved cytosolic proteins that catalyze the peptidyl-prolyl cis-trans isomerazation reaction before protein folding process, it has been suggestive of an allergen in a few numbers of fungi such as aspergillus fumigatus and malassezia species. allergenic cyclophilins are ige-binding components, which have been characterized in other species of malassezia ; and are considered as mala s 6 in malassezia sympodialis .  in the present study we tried to identify the molecular characterization of cyclophilin gene in m. furfur. methods: pairs of oligonucleotide primers were designed from highly conserved regions of the gene counterparts in other fungi. the primers were then applied to amplify the primer-specific dna fragment. afterward, pcr product fragments were sequenced to be used in further analysis. results: about 573 nucleotides, encoding a polypeptide of 190 amino acids, have been sequenced. sequence comparison was performed in gene bank, both for the nucleotides and their deduced amino acid sequence. it revealed a significant homology with cyclophilin genes and proteins of other eukaryotic cells. the amino acid sequence of the encoded protein was about 86% identical to the sequence of cyclophilin protein from other fungi. conclusion: the molecular characterization of cyclophilin gene may open the way to disclosure of the functional characteristics of cyclophilin and is a fundamental step for understanding the molecular basis of its pathogenesis in aeds disease.

Malassezia Furfur in Malaysia

An in-house prepared M. furfur antigen was used to carry out a seroprevalence study in an urban population in Malaysia by indirect immunofluorescence assay. Of the 800 serum samples from all ages screened, 738 samples were positive for M. furfur specific IgG, giving an overall seropositive rate of 92.3%. There was no significant difference in the seropositive rates among the different gender gr...

Molecular analysis of malassezia microflora on the skin of the patients with atopic dermatitis.

BACKGROUND The yeasts of the genus Malassezia are members of the normal flora on human skin and they are found in 75~80% of healthy adults. Since its association with various skin disorders have been known, there have been a growing number of reports that have implicated Malassezia yeast in atopic dermatitis (AD). OBJECTIVE The aim of the present study is to isolate the various Malassezia spe...

Rapid Identification of Malassezia furfur from other Malassezia Species: A Major Causative Agent of Pityriasis Versicolor

The present study is designed to evaluate the application of a simple method for rapid identification of Malassezia furfur among other Malassezia spp. based on production of a brownish-red pigment and its diffusion into the medium containing L-tryptophan as a sigle source of nitrogen.  91 strains of Malassezia (20% M. furfur, 2% M. sympodialis, 5% M. obtosa and 73% M. globosa) isolated from ski...

Different utilization of neutral lipids by Malassezia furfur and Malassezia sympodialis.

In recent years, the genus Malassezia has been expanded based on molecular data; in addition to M. furfur and M. pachydermatis, five new species (M. sympodialis, M. globosa, M. obtusa, M. restricta, M. slooffiae) have been described. Apart from their lipid dependence, little is known about the metabolism and nutritional requirements of these new species. Defined inocula of Malassezia reference ...
